Comparison of the inhibition of TC uptake by bosentan in human and rat hepatocyte cultures and HepaRG cell cultures.

(A) HepaRG cells were treated with 1 µM of ³H-TC for 300 sec at 4, 24 and 96 h after seeding in the presence of bosentan (10–200 µM), or the vehicle, respectively. (B) Human and (C) rat hepatocytes were treated with 1 µM of ³H-TC for 30 sec at 4, 24 and 72 h (rat) or 4, 120 h (human) after seeding in the presence of bosentan (10–100 µM), or the vehicle, respectively. Values are expressed as mean±SD, assays were run using four wells as one set. All experiments were carried out with hepatocytes from three independent preparations.
